    Knowledge Acquisition Analytical Games: games for cognitive systems design

    Knowledge discovery from data and knowledge acquisition from experts are steps of paramount importance when designing cognitive systems. The literature discusses extensively on the issues related to current knowledge acquisition techniques. In this doctoral work we explore the use of gaming approaches as a knowledge acquisition tools, capitalising on aspects such as engagement, ease of use and ability to access tacit knowledge. More specifically, we explore the use of analytical games for this purpose. Analytical game for decision making is not a new class of games, but rather a set of platform independent simulation games, designed not for entertainment, whose main purpose is research on decision-making, either in its complete dynamic cycle or a portion of it (i.e. Situational Awareness). Moreover, the work focuses on the use of analytical games as knowledge acquisition tools. To this end, the Knowledge Acquisition Analytical Game (K2AG) method is introduced. K2AG is an innovative game framework for supporting the knowledge acquisition task. The framework introduced in this doctoral work was born as a generalisation of the Reliability Game, which on turn was inspired by the Risk Game. More specifically, K2AGs aim at collecting information and knowledge to be used in the design of cognitive systems and their algorithms. The two main aspects that characterise those games are the use of knowledge cards to render information and meta-information to the players and the use of an innovative data gathering method that takes advantage of geometrical features of simple shapes (e.g. a triangle) to easily collect players\u2019 beliefs. These beliefs can be mapped to subjective probabilities or masses (in evidence theory framework) and used for algorithm design purposes. However, K2AGs might use also different means of conveying information to the players and to collect data. Part of the work has been devoted to a detailed articulation of the design cycle of K2AGs. More specifically, van der Zee\u2019s simulation gaming design framework has been extended in order to account for the fact that the design cycle steps should be modified to include the different kinds of models that characterise the design of simulation games and simulations in general, namely a conceptual model (platform independent), a design model (platform independent) and one or more implementation models (platform dependent). In addition, the processes that lead from one model to the other have been mapped to design phases of analytical wargaming. Aspects of game validation and player experience evaluation have been addressed in this work. Therefore, based on the literature a set of validation criteria for K2AG has been proposed and a player experience questionnaire for K2AGs has been developed. This questionnaire extends work proposed in the literature, but a validation has not been possible at the time of writing. Finally, two instantiations of the K2AG framework, namely the Reliability Game and the MARISA Game, have been designed and analysed in details to validate the approach and show its potentialities

    In this essay I will discuss the iconographic constructions developed by the Portuguese colonial project, arguing that Estado Novo has used cinema to consolidate social categories defined by the regime propaganda, using a discourse based on reality and authenticity and through the projection of stereotyped structures such as race and gender. Moving in the background of the theoretical frame on the concepts of archive and digital archive, the analysis will focus on a deconstructive re-interpretation of colonial and dominant narratives in two documentaries realized for the Portuguese industrial exposition in 1932, África em Lisboa – Os Indígenas da Guiné na Grande Exposição Industrial ((Africa in Lisbon- The Indigenous People of Guinea in The Great Industrial Exhibition) and Guiné Aldeia Indígena em Lisboa – 1932 (Guinea Indigenous Village in Lisbon- 1932). The discussion will hence be centered on the representation and the construction of relations of dominance and power over the black female body

    Direct conversion of human pluripotent stem cells into cranial motor neurons using a piggyBac vector

    Human pluripotent stem cells (PSCs) are widely used for in vitro disease modeling. One of the challenges in the field is represented by the ability of converting human PSCs into specific disease-relevant cell types. The nervous system is composed of a wide variety of neuronal types with selective vulnerability in neurodegenerative diseases. This is particularly relevant for motor neuron diseases, in which different motor neurons populations show a different susceptibility to degeneration. Here we developed a fast and efficient method to convert human induced Pluripotent Stem Cells into cranial motor neurons of the branchiomotor and visceral motor subtype. These populations represent the motor neuron subgroup that is primarily affected by a severe form of amyotrophic lateral sclerosis with bulbar onset and worst prognosis. This goal was achieved by stable integration of an inducible vector, based on the piggyBac transposon, allowing controlled activation of Ngn2, Isl1 and Phox2a (NIP). The NIP module effectively produced electrophysiologically active cranial motor neurons. Our method can be easily extended to PSCs carrying disease-associated mutations, thus providing a useful tool to shed light on the cellular and molecular bases of selective motor neuron vulnerability in pathological conditions

    Multidisciplinary Approach to Structural/Energy Diagnosis of Historical Buildings: A Case Study

    Abstract A synergic approach for the investigations of the historical building performances - with reference to both the structural behavior and the energy performances for the space heating and cooling - is presented. The historical masonry building "Palazzo Bosco Lucarelli", located in Benevento, has been chosen as case study. The structural and energy analyses are carried out in parallel, especially during the identification of the building characteristics through tests and surveys in-situ. For the structural analysis - beyond examinations on materials - some dynamic tests have been used for better assessing a numerical Finite Element model necessary for the verification of the structure safety. Moreover, being necessary a structural refurbishment, also an energy retrofit could be realized. A rigorous evaluation procedure - aimed to guarantee the necessary reliability of numerical predictions - is performed in order to verify the technical and economical convenience of various energy retrofit solutions

    Physical education teacher training for disability

    This work in focused on investigation of physical education teachers’ education with pedagogical bases for students with disabilities. Starting from an American research on teachers of physical education who work in contact with the disability, it has shown how the teachers express negative attitudes towards inclusion because they consider their training inadequate. The aim is to understand the phenomenon of physical education teacher training analysing the documentary research of the master’s degree programs in sports sciences in Italy, with a specific study on the master’s degree in sports sciences at the university of Salerno and, finally, a survey on teachers’ perception of disability. The first study revealed that the main difficulty encountered is the inability to plan inclusive and personalized lessons, since the degree courses refer to the bio-medical area. The second study, the analysis of the Master degree course of Science in Sports Science at the University of Salerno, mainly refers to the pedagogical-didactic area and aimed at teaching and designing for the purpose of education for the disabled. From the fact-finding survey on teachers’ perceptions emerges that both pedagogy and teaching are fundamental for any job in the motor and sports field, even for those who decide not to teach. In conclusion, the training of the physical education teacher who also teaches the disabled must have a less bio-medical and more pedagogical education

    High cell density cultivation of Escherichia coli K4 in a microfiltration bioreactor: a step towards improvement of chondroitin precursor production

    <p>Abstract</p> <p>Background</p> <p>The bacteria <it>Escherichia coli </it>K4 produces a capsular polysaccharide (K4 CPS) whose backbone is similar to the non sulphated chondroitin chain. The chondroitin sulphate is one of the major components of the extra-cellular matrix of the vertebrate connective tissues and a high value molecule, widely employed as active principle in the treatment of osteoarthritis. It is usually obtained by extraction from animal tissues, but the risk of virus contaminations, as well as the scarceness of raw material, makes this productive process unsafe and unable to satisfy the growing market demand. In previous studies a new biotechnological process to produce chondroitin from <it>Escherichia coli </it>K4 capsular polysaccharide was investigated and a 1.4 g·L<sup>-1 </sup>K4 CPS concentration was reached using fed-batch fermentation techniques. In this work, on the trail of these results, we exploited new fermentation strategies to further improve the capsular polysaccharide production.</p> <p>Results</p> <p>The inhibitory effect of acetate on the bacterial cells growth and K4 CPS production was studied in shake flask conditions, while a new approach, that combined the optimization of the feeding profiles, the improvement of aeration conditions and the use of a microfiltration bioreactor, was investigated in three different types of fermentation processes. High polysaccharide concentrations (4.73 ± 0.2 g·L<sup>-1</sup>), with corresponding average yields (0.13 ± 0.006 g<sub>K4 CPS</sub>·g<sub>cdw</sub><sup>-1</sup>), were obtained; the increase of K4 CPS titre, compared to batch and fed-batch results, was of 16-fold and 3.3-fold respectively, while average yield was almost 3.5 and 1.4 fold higher.</p> <p>Conclusion</p> <p>The increase of capsular polysaccharide titre confirmed the validity of the proposed fermentation strategy and opened the way to the use of the microfiltration bioreactor for the biotechnological production of chondroitin.</p

    CHM/REP1 Transcript Expression and Loss of Visual Function in Patients Affected by Choroideremia

    PURPOSE. To evaluate the disease progression in patients with clinical and genetic diagnoses of choroideremia during a long-term follow-up and to investigate the relationship between pathogenic variants in the CHM/REP1 gene and disease phenotypes. METHODS. We performed a retrospective longitudinal study on 51 affected men by reviewing medical charts at baseline and follow-up visits to extract the following ocular findings: best-corrected visual acuity, Goldmann visual field, optical coherence tomography, microperimetry. Data obtained from the analysis of DNA and mRNA were reevaluated for genetic classification of patients. RESULTS. The longitudinal analysis showed a significant (P < 0.001) worsening of best-corrected visual acuity with a mean rate of 0.011 logMar per year before 50 years and 0.025 logMar per year after 50 years. Similarly, V4e Goldmann visual field area significantly (P ≤ 0.01) decreased at a mean rate of 2.7% per year before 40 years and 5.7% after 40 years. Moreover, we observed a significant (P < 0.05) decrease of macular sensitivity with a mean rate of 5.0% per year and a decrease of mean macular thickness with a mean rate of 0.8% per year. We classified our patients into two groups according to the expression of the CHM/ REP1 gene transcript and observed that mutations leading to mRNA absence are associated with an earlier best-corrected visual acuity and Goldmann visual field loss. CONCLUSIONS. Our analysis of morphological and functional parameters in choroideremia patients showed a slow disease progression, particularly in the first decades of life. Overall, reevaluation of clinical and molecular data suggests exploring the genotype–phenotype relationship based on CHM/REP1 transcript expression. PURPOSE.     Design methodology of analytical games for knowledge acquisition

    Analytical games explore a problem or a domain with a research purpose. Considerable research is ongoing to investigate improvements to analytical game design, execution and exploitation. Moreover, the fast-paced technological developments in many fields, such as artificial intelligence and virtual reality, make it even more compellingto account for the advantages and limitations of these new capabilities. In game design, the use of digital means is often regarded as a mere technical factor that relates to the platform selection, facilitator support and data recording processes. In this work a shift in perspective is proposed, to move from technology-oriented design selection criteria towards a broader assessment of the design choices. In fact, the introductionof technology (i.e., automation and autonomy) will not lead to a substitution of tasks, but will intrinsically change the game environment. This work introduces a framework to provide a structured guidance on the aspects to be factored in the different design phases of an analytical game, including the potential impact of the adoption of automation and autonomy. The proposed approach is based on previous research in the fieldof simulation-based serious gaming, model-driven engineering and human factors engineering.
